Shootは「撃つ」という動詞ですが、この慣用句では名詞shotを使います。Shotには一発の射撃という意味のほか、比喩的に一度の試みという意味があります。すでにgiveという動詞があり、aの後には名詞が必要なので、a shotとします。
Give it a shot、give it a try、英国英語のgive it a goは、ほぼ同じ意味です。一方give it your best shotは、できる限り力を尽くすことです。The exam is hard, but just give it your best shot.のように、難しいことに向かう人を励ますときに使います。
