Line up meaning: to queue, or to arrange

To stand in a queue or a row. But a colleague who says I've got a new job lined up isn't queuing for anything: they're ahead of everyone. What did they just tell you?

Line up means to stand in a row or join a queue: line up for tickets. The second meaning is to arrange something in advance: line up a speaker, have a job lined up. The last one means the job has already been found and agreed, so there's nothing left to look for.
